Asparagine biosynthesis in human KB tumor cells: inhibitor studies with asparagine and glutamine antagonists.

چکیده
SUMMARY Resistance to L-asparagine depletion in tumor cells has been correlated with asparagine biosynthesis mediated by the enzyme asparagine synthetase. In this study , we describe certain characteristics of the asparagine synthetase of the KB human tumor cell carried in tissue culture. The KB enzyme is similar to that of other mammalian tumors in terms of c ofactor requirements and inhibition by L-asparagine antagonists. The in vitro effect of glutamine antagonists on enzyme activity was determined. 6-Diazo-5-oxo-L-norleucine and azotomycin were shown to be effective inhibitors, but azaserine was relatively ineffective. The implications of combining such drugs with the antineoplastic agent L-asparaginase are discussed.

Changes of glutamine and asparagine content in cucumber seedlings in response to nitrate stress
Nitrogen fertilizer application rates in intensive agricultural systems have increased dramatically in recent years, especially in protected vegetable production systems. This excessive use of nitrogen fertilizer has resulted in soil secondary salinity, which has become a significant environmental stress for crops such as cucumber, in the protected farmlands. It is thus necessary to illuminate ...
متن کاملA Critical Role of Glutamine and Asparagine γ-Nitrogen in Nucleotide Biosynthesis in Cancer Cells Hijacked by an Oncogenic Virus
While glutamine is a nonessential amino acid that can be synthesized from glucose, some cancer cells primarily depend on glutamine for their growth, proliferation, and survival. Numerous types of cancer also depend on asparagine for cell proliferation. The underlying mechanisms of the glutamine and asparagine requirement in cancer cells in different contexts remain unclear. In this study, we sh...
متن کاملSynthetic lethality of glutaminolysis inhibition, autophagy inactivation and asparagine depletion in colon cancer
Cancer cells reprogram metabolism to coordinate their rapid growth. They addict on glutamine metabolism for adenosine triphosphate generation and macromolecule biosynthesis. In this study, we report that glutamine deprivation retarded cell growth and induced prosurvival autophagy. Autophagy inhibition by chloroquine significantly enhanced glutamine starvation induced growth inhibition and apopt...
متن کاملThe effect of asparagine and adenine on the glutamine requirement for growth of human peripheral lymphocytes.
Quantitative growth responses of lymphocytes directly isolated from individual subjects in a newly developed chemically-defined, protein-free medium are used to demonstrate that supplements of both L-asparagine and a purine source, but neither alone, significantly reduce the quantitative requirement for L-glutamine for growth. This system is useful for exploring individual differences in quanti...
متن کاملThe Utilization of Glutamine, Glutamic Acid, and Ammonia for the Biosynthesis of Nucleic Acid Bases in Mammalian Cell Cultures* by Norman
Glutamine has proved to have multiple metabolic functions in tissue cultures of animal and human cells. It is directly incorporated into protein; and in a limiting medium it is used for the biosynthesis of glutamic acid, aspartic acid, and proline (2), as well as asparagine (3). The present studies were undertaken to clarify the role of glutamine in the biosynthesis of the nucleic acid bases. I...
